FOR PATIENT FRIENDLY DELIVERY OF CHALLENGING POLYFUNCTIONAL DRUGS The development of successful parenteral products, especially IV formulations, often is hampered by challenges associated with the solubilization of actives at the appropriate therapeutic amounts within a given unit dosage form. Conventional solubilization approaches of IV formulations have the potential for irritation, anaphylactic response, necrosis, and/or volume doping. Other demanding aspects of IV formulations, for example, include compatibility, compliance, and economic issues of the components or methods adopted for the solubilization of such actives. LiIV entails innovative lipid compositions designed to provide adequate loading capacities for hydrophobic actives that remain solubilized upon infusion. LiIV compositions are customized to match closely the polarities of the lipid vehicles with those of the target actives. LiIV formulations are significantly free of organic solvents or surfactants that attribute to side effects found using conventional parenteral formulations. LiIV compositions enable lipid carriers to insulate harmful or irritating side effects of actives caused by direct contacts between actives and tissues and/or organs at the site of administration, during and/or following the administration. LiIV compositions also can be coupled with other actives or passive targeting approaches for advanced drug delivery. Overall, LiIV-based formulations are designed to have an improved therapeutic index of actives compared to conventional parenteral dosage forms.
